Using Exchange 5.5 on the back-end, I can't remember how to set up conference
rooms as a resource and to be able to schedule them.

Hi Sue, I'm very close and setup the mailbox as a Direct Booking then logged
into the mailbox and created an item, set permission to Author for the
company.  Then tried to book it as a resource from my system and I get the
following error message back: "The calendar folder could not be found. Unable
to directly book a resource for this meeting".  Any advice and thanks in
advance...

Can you open it with the FIle | Open | Other User's Folder command?

What about the name of the room? Is it possible that you're typing in a name
that's resolving to something other than the mailbox?

You did set Author permission on the Calendar folder, right?
