Can I change the default meeting response to "Don't Send.."?

Is there a way to change the default response, when accepting a meeting
request, to "Don't Send a Response"? Most of my meeting requests are for
large groups of attendees, and the meeting requestor asks that we don't send
a response.

Not that I can see. Outlook (assuming the meeting request comes from
Outlook) needs that feedback to update the Meeting info.
If the Organizer doesn't want a Response I suggest they just email an
Appointment.
