Not sure if I can pose this question simply or not, but here goes:
I would like to set a "Response Reminder" on an email I send to someone so
that if they do not answer me back in say two days, that I get an automatic
reminder to send them another email or call them on the land line to find out
why they have not responded.
This provides a couple of functions, first if the message does not reach
them (bad email address) it reminds me to seek other methods to contact them,
Second and more importantly, if they do not handle a specific job I want done
and answer me back by the time I set, then I can go and chew some tail.
Does this make sense?

There's no way to make Outlook trigger on a non-event; i.e., you can't set a
reminder for something that doesn't happen. Outlook contains no per-message
or per-recipient timers.

you can flag messages in the sent folder - it won't tell you if you received
a reply, just remind you to look for a reply. There are also 3rd party tools
that will remind you - such as follow up reminder on this page:
