Hi,
I have created a template with new toolbars and menu items
which rolls out firm wide, called firm.dot. I want to
make a custom label definition and have it rolled out to
all users via the firm.dot. However, when I open the
firm.dot to create the custom label, it is not saved to
the firm.dot in the same way I have been able to save
menus, buttons, etc.
How can I save a custom label definition to a firm.dot
template?

Custom label definitions are saved in the Registry, not in Normal.dot.

You can export the registry key
HKEY_CURRENT_USER\Software\Microsoft\Office\11.0\Word\Custom Labels
and users could import the resulting file into their registries, provided
they have the same Word version.
