There is no setting for this, because Word is a wordprocessor, not a
typewriter. It uses proportional fonts and thus double spaces are
irrelevant. If you insist the only way is to use the replace function.

You can actually specify that the Spelling and Grammar tool underline
anything other than two spaces so you can see when you've forgotten:
1. Tools > Options > Spelling and Grammar
2. Click the Settings button for Grammar (beneath writing style)
3. In the Require section, the 3rd option is for the number of spaces
between sentences. Set this to 2 and click OK.
4. Click OK in the Options dialog box.

Unless you are using monospaced fonts (such as Courier New) to emulate a
typewriter, you don't want two spaces after periods (or colons or question
marks). The spacing for these is built into proportionally spaced fonts.
